Shaeffer and Fulton modified Dorner’s method in 1933 to make the process faster The endospore stain is a differential stain which selectively stains bacterial endospores. The main purpose of endospore staining is to differentiate bacterial spores from other vegetative cells and to differentiate spore formers from non-spore formers.

Endospore Stain Endospores are dormant forms of living bacteria and should not be confused with reproductive spores produced by fungi. These structures are produced by a few genera of Gram-positive bacteria, almost all bacilli, in response to adverse environmental conditions. Two common bacteria that produce endospores are Bacillus or Clostridum.

Endospore staining is a unique staining method, which discriminates between the two major groups of bacteria (spore formers and non-spore formers) by selectively staining the endospore against the vegetative cell. Clostridium perfringes, Bacillus cereus, Sporosarcina sp etc., are examples of endospore formers. This method, called the endosporestain, uses either heat or long exposure time to entice the endospores to take up the primary stain, usually a water soluble dye such as malachite green since endospores are permeable to water. Endospore Staining is a technique used in bacteriology to identify the presence of endospores in a bacterial sample, which can be useful for classifying bacteria.

A primary stain in the form of malachite green is used for staining endospores. It uses the heating process to act as a mordant as endospores have the ability to resist staining. Such a procedure does not require the use of decolorizer because malachite green binds to the cell and spore wall.

Ø The classically used stain to visualize endospore is Malachite Green and the staining procedure is known as Schaeffer–Fulton Staining. Ø The endospore-producing mother cell is called sporangium. Endospores are quite resistant to most staining procedures; however, in a routinely stained smear, they may be visible as “outlines” with clear space within.
